چکیده
Objective: The goal of this study was to determine whether PDE4D regulates basal cardiac contractility. Methods and Results: At 10 to 12 weeks of age, baseline cardiac contractility in PDE4D-deficient (PDE4D / ) mice was elevated mice in vivo and in Langendorff perfused hearts, whereas isolated PDE4D / cardiomyocytes showed increased whole-cell Ca transient amplitudes and SR Ca content but unchanged

Ca 2 Scraps Local Depletions of Free [ Ca 2 ] in Cardiac Sarcoplasmic Reticulum
Free [Ca ] inside the sarcoplasmic reticulum ([Ca ]SR) is difficult to measure yet critically important in controlling many cellular systems. In cardiac myocytes, [Ca ]SR regulates cardiac contractility. We directly measure [Ca ]SR in intact cardiac myocytes dynamically and quantitatively during beats, with high spatial resolution. Diastolic [Ca ]SR (1 to 1.5 mmol/L) is only partially depleted ...

متن کاملCellular Mechanisms of Altered Contractility in the Hypertrophied Heart
To investigate the cellular mechanisms for altered Ca homeostasis and contractility in cardiac hypertrophy, we measured whole-cell L-type Ca currents (ICa,L), whole-cell Ca 21 transients ([Ca]i), and Ca 21 sparks in ventricular cells from 6-month-old spontaneously hypertensive rats (SHRs) and from ageand sex-matched Wistar-Kyoto and Sprague-Dawley control rats. By echocardiography, SHR hearts h...
متن کاملHeart Failure Cardiac G-Protein–Coupled Receptor Kinase 2 Ablation Induces a Novel Ca Handling Phenotype Resistant to Adverse Alterations and Remodeling After Myocardial Infarction
cardiomyocytes from wild-type and GRK2 knockout (GRK2KO) mice without (sham) or with myocardial infarction (MI). In cardiac myocytes isolated from unstressed wild-type and GRK2KO hearts, myocyte contractions and Ca transients were similar, but GRK2KO myocytes had lower sarcoplasmic reticulum (SR) Ca content because of increased sodium-Ca exchanger activity and inhibited SR Ca ATPase by local pr...
